Study on biochemical phenotypic changes and heterogeneity of macrophages after phagocytosis of myelin fragments

Based on a cell model of macrophages phagocytosis of myelin fragments, synchrotron radiation Fourier transform infrared (SR-FTIR) spectroscopy was used to successfully characterize biochemical changes in macrophages at the single-cell level. Through the calculation and statistical analysis of infrared spectrum fluctuation, principal component analysis, and Euclidean distance between cells, this study obtained the exact evidence of the simultaneous dynamic changes of lipids and proteins in macrophages after phagocytic myelin fragments, which highlights the potential of SR-FTIR to provide biochemical macromolecule information at the single-cell level.
