Applications of infrared phenomics in microbiology

Biofilm infection can cause long-term and chronic inflammation, and it is extremely difficult to cure. Therefore, it is urgent to develop methods for in-depth characterization of complex biofilm phenotypes to reveal its most basic biological characteristics and clinical treatment.

This work developed a method to measure biofilm phenotype based on infrared microspectroscopy, which could quantitatively evaluate biofilm heterogeneity based on the similarity between infrared spectral data, and then revealed the phenotypic differences between two E. coli strains during biofilm formation. Combined with two-dimensional correlation spectroscopy (2D-COS), the biochemical molecular evolution sequence of E. coli biofilm formation was further explored, and polysaccharide components were found to change first, expanding the new application field of infrared microspectroscopy to reveal the molecular evolution rule of biofilm formation.
